Studying RNA Structural Heterogeneity Using Direct RNA Sequencing

RNA structure can determine RNA function. However, RNAs usually do not fold into only one conformation in solution or inside cells. To study the complexity of RNA structures, we utilize SHAPE chemical probing with nanopore direct RNA sequencing to read out RNA structural information along single molecules. We show that the approach works on known RNA switches and applied the method to identify heterogenous regions along SARS-CoV-2 and candida albicans.
