TNF Signatures Identified Based on Integrating Transcriptomics and Proteomics and the Application in Diseases

Tumour necrosis factor (TNF) is an important cytokine in inflammation and cell death, that can induce different outcomes, including inflammation signalling, apoptosis, and necroptosis in cells. Its critical role in disease progression makes it a big clinical target and research focus. Anti-TNF therapies have been used in inflammatory diseases for decades with large consumption yearly. While in cancers, the TNF signalling has interestingly conflicting functions according to disease type and conditions, with people making efforts to turn it into drugs. There have been several predictive or prognostic TNF molecular signatures found separately in each single disease and using single omics, with a lack of general view combing multiple omics or tissues. In my work, by integrating transcriptomics and proteomics to study TNF signalling in multiple cell lines, I identified transcriptomic TNF signatures broadly expressed in multiple cell lines and an endothelial signature with both transcriptomic and proteomic evidence. The signature shows high correlation with survival in TCGA patients and anti-TNF treatment in Rheumatoid arthritis patients.
