Digital Spatial Profiling transcriptomics and proteomics analyses

In recent years, new spatial profiling technologies (such as NanoString GeoMx Digital Spatial Profiling) that leverage on multiplexed labeling are enabling the detection of molecular biomarkers in their native spatial state. These technologies along with the development of computational analysis methods have started to drive more comprehensive investigations and provide new insight into disease pathogenesis and tumor heterogeneity not only in cancer but across other diseases. In this talk I will discuss two such collaborative projects.

The first is a COVID-19 lung digital spatial transcriptomics project which uses a bioinformatics approach to identify candidate novel biomarkers and pathways involved in viral infections specific to COVID-19. An in-depth picture of the pulmonary transcriptional landscape for SARS-CoV2 infection, H1N1 influenza infection and uninfected control patients were generated by collecting and profiling lung biopsies. This analysis offers new insights into the pathogenesis of SARS-COV-2 virus and aids in patient triage and treatment.

The second project uses digital spatial protein profiling to examine the immune microenvironment in Hairy Cell Leukaemia pre- and post-therapy. A detailed analysis of the bone marrow microenvironment was conducted to determine potential biomarkers of durable response to therapy that would guide identification and management of patients with a high risk of relapse post-therapy.
