Where have I put that gene? Genomic integration site analysis for biopharmaceutics applications

Delivery of designer genes into a host genome plays a critical role in the biopharmaceutic process. Let it be transfecting genes into a cell line for the production of recombinant protein or monoclonal antibodies; or transducing therapeutic genes into patients’ cells to restore defective function.

Currently, the delivery method is largely uncontrolled, either by design or as the result of technology of choice. Determining where and how the vector has been integrated into the genome helps to understand the safety, productivity and effectiveness of the resultant product.

In this talk, I will highlight the importance of identifying genomic integration sites in the biopharmaceutics context; speak to our continued effort in exploring genomic methods for characterizing the product genome for integration site; and then focus on our experiences of using two niche and bespoke methods of sequencing for this purpose: Targeted Locus Amplification and Cas9-enriched Nanopore sequencing.
