Long-Range Methlation from Nanopore Sequencing

Chris Woodruff
WEHI Bioinformatics

Methylation along the genome has been shown to be correlated over ranges of up to 1000 bases. the strength of correlation reflects known DNA conformational features such as helical periodicity and nucleosomal structure. We have used long-read DNA sequencing to extend this analysis over a 50x greater range. This talk will outline recent prior work on comethylation ; how nanopore sequencing allows methylation determination ; and analysis of long range comethylation on the mouse genome. Some preliminary consideration of how this might relate to 3D conformation of genomic material will also be presented.
