Nanopore Sequencer Data Analysis - facts

fictions ; and caveats;Nanopore sequencing is highly topical ; potentially extremely valuable ; but - for good reasons - not yet a commercialised technology. Over the past 12 months we have been generating data from an Oxford Nanopore Technologies minION sequencer. Initially we focussed on the basecalling performance on E.Coli. More recently we have been looking at the latest developments in identifyingDNA based modifications ; and we are moving to be able to investigate possible solutions to the limitations of the minION in handling homopolymers. In this talk I will discuss the nature of the data that the nanopore sequencer provides ; how basecalling is achieved with this data ; recent published material on recognising base modifications both in RNA and DNA ; and highlight the difficulties and possible solutions of homopolymer calling. Finally a brief comparison with Pacbio technology and performance will be provided ; focussed on recognition and localisation of DNA modifications;
