Gene expression analysis and expression QTL analysis: application to the human brain

Over the past decade ; there have been tremendous improvements in genetic technologies allowing researchers to obtain information at the whole-genome level. This has led to new opportunities to understand the genetic architecture ; in animal ; plant ; and human systems. One of the challenges however is to develop appropriate analysis methodologies to cope with these high-dimensional data. This talk will describe an approach we have developed for analysing gene expression in multiple regions of the human brain. A two-stage approach is used ; firstly using a large-scale mixed model to analyse all expression data simultaneously ; followed by use of a finite mixture model to determine differentially expressed genes. Using this approach ; patterns of multi-region gene expression have been investigated. Recently ; the investigation has been extended into expression QTL (eQTL) through use of SNP arrays in conjunction with the expression arrays. Some details of approaches and results will be discussed.
